5-Azacitidine, chemically known as 1-(beta-D-ribofuranosyl)-4-amino-1,3,5-triazin-2(1H)-one, also known as 5-azacytidine, azacitidine, Vidaza, is a white needle-like crystal; it is a DNA methyltransferase inhibitor developed by Pharmion, Inc. and first marketed in the United States in July 2004. It is a cell cycle-specific drug that acts in the S phase and rapidly phosphorylates and penetrates into RNA (ribonucleic acid) and DNA (deoxyribonucleic acid) to inhibit protein synthesis by disrupting the smooth translation of nucleic acids into proteins, and also affects the synthesis of pyrimidines by inhibiting orotate nucleotide decarboxylase. It is used clinically to treat myelodysplastic syndromes including refractory anaemia, refractory anaemia with cyclic iron granulocytosis, if accompanied by neutropenia, thrombocytopenia or requiring blood transfusions, refractory anaemia with primocytosis, refractory anaemia with primocytosis-transformed and five subtypes of chronic myelomonocytic leukaemia.

1. 5-Azacytidine, its incorporation into RNA alters RNA synthesis and processing, and results in inhibition of protein synthesis. It has been used as a cancer chemotherapeutic agent. It is a powerful bacteriostatic, antitumor, and mutagenic agent; it also exhibits immunosuppressive, antimitotic, radioprotective, and virostatic effects.
2. 5-Azacytidine is a potent growth inhibitor and cytotoxic agent. It acts as a demethylating agent by inhibiting DNA methyltransferase
3. 5-Azacytidine is used to treat the following subtypes of myelodysplastic syndrome: refractory anaemia (RA), circumferential iron-granulocytic refractory anaemia (RARS) (with neutropenia or thrombocytopenia or requiring blood transfusion), refractory anaemia with primocytosis (RAEB), refractory anaemia with primocytosis transformed (RAEB-T) and chronic granulocytic mononuclear leukaemia (CMMoL).
